چکیده

Aequorin is a Ca-sensitive photoprotein isolated from the jellyfish Aequorea victoria. It is an ideal probe for measuring Ca concentration ([Ca]) in intracellular organelles because it can be modified to include specific targeting sequences. On the binding of Ca to three high-affinity sites in aequorin, an irreversible reaction occurs in which the prosthetic group coelenterazine is released and a photon is emitted. This protocol presents procedures for expressing, targeting, and reconstituting aequorin in intact and permeabilized mammalian cells and describes how to use this photoprotein to measure intracellular [Ca] in various subcellular compartments.

Using targeted variants of aequorin to measure Ca2+ levels in intracellular organelles.

Aequorin is a Ca(2+)-sensitive photoprotein isolated from the jellyfish Aequorea victoria. It is an ideal probe for measuring Ca(2+) concentration ([Ca(2+)]) in intracellular organelles because it can be modified to include specific targeting sequences. On the binding of Ca(2+) to three high-affinity sites in aequorin, an irreversible reaction occurs in which the prosthetic group coelenterazine...

Ca2+ Measurements in Mammalian Cells with Aequorin-based Probes.

Aequorin is a Ca2+ sensitive photoprotein suitable to measure intracellular Ca2+ transients in mammalian cells. Thanks to recombinant cDNAs expression, aequorin can be specifically targeted to various subcellular compartments, thus allowing an accurate measurement of Ca2+ uptake and release of different intracellular organelles. Here, we describe how to use this probe to measure cytosolic Ca2+ ...

GFP-Aequorin Protein Sensor for Ex Vivo and In Vivo Imaging of Ca(2+) Dynamics in High-Ca(2+) Organelles.

Proper functioning of organelles such as the ER or the Golgi apparatus requires luminal accumulation of Ca(2+) at high concentrations. Here we describe a ratiometric low-affinity Ca(2+) sensor of the GFP-aequorin protein (GAP) family optimized for measurements in high-Ca(2+) concentration environments. Transgenic animals expressing the ER-targeted sensor allowed monitoring of Ca(2+) signals ins...

Utilizing Hidex Sense for mitochondrial and cytosolic calcium measurements using the luminescent calcium reporter protein aequorin and the fluorescent calcium indicator Fura-2 AM

reporter. Aequorin is a Ca 2+ -sensitive luminescent protein originally derived from the jellyfish Aequorea victoria. When expressed in mammalian cells, wild-type aequorin localizes to the cytoplasm and can be employed for cytoplasmic Ca 2+ measurements. Recombinant aequorin fusion proteins with specific targeting peptide sequences have been produced in order to direct the reporter protein to a...

Bioluminescence imaging of calcium oscillations inside intracellular organelles.

Ca(2+) oscillations inside intracellular organelles are important for regulation of functions such as gene expression at the nucleus, respiration at mitochondria or protein processing at the endoplasmic reticulum. Targeted aequorins are excellent calcium probes for subcellular analysis, but single-cell imaging has proven difficult because of low light yield. Here we describe a procedure that co...
